The aim of this study was to compare cytotoxic potential of ambient air particulate matter PM10 (< 10 μm) sampled in February/March 2018 in 4 different sampling sites (Lom u Mostu, Czech Republic, Deutschneudorf, Collmberg, Leipzig Mitte, all Germany) in model of the human lung cells A549 with the data of the campaign accomplished in February/March 2017. As in the previous study the organic components of PM10 collected in 4 localities were extracted and cytotoxicity of extracts was tested at concentrations 1, 5, 15 a 30 m3 of sampled air/ml using WST-1 cytotoxicity assay. In contrast to 2017 (10 – 38 μg/m3 in Lom u Mostu, 3 – 15 μg/m3 in Deutschneudorf, 3 – 13 μg/m3 in Collmberg, 11 – 33 μg/m3 in Leipzig Mitte), in 2018 daily PM10 levels from all 4 sampling stations exceed in some extent the daily PM10 limit value of 50 μg/m3 and varied between 11.5 - 83.4 μg/m3 (Lom u Mostu), 16.8 – 57.4 μg/m3 (Deutschneudorf), 12.9 - 75.1 μg/m3 (Leipzig Mitte), 8.4 - 50.5 μg/m3 (Collmberg). Similar to 2017 cytotoxicity of PM10 from all localities, cytotoxicity of PM10 sampled in 2018 is generally low and depending on the locality cell survival varied between 75 – 90 % for highest PM10 concentration tested. The comparison of cytotoxic effect between localities as measured by air volume needed to kill 30 % cells (LC30 = lethal concentration killing 30 % cells) suggests following order: Lom u Mostu (LC30=36 m3 of sampled air/ml) > Deutschneudorf (LC30=45 m3/ ml) > Leipzig Mitte (LC30=53 m3/ml) > Collmberg (LC30=76 m3/ml). As in 2017 (Lom u Mostu LC30=28 m3/ml > Leipzig Mitte LC30=40 m3/ml > Deutschneudorf LC30=53 m3/ml >> Collmberg LC30=116 m3/ml) PM10 from Lom u Mostu remained the most cytotoxic in 2018, while Collmberg exhibited lowest PM10 toxicity. Generally, the differences in toxic potential among localities are smaller than in 2017. Toxicity did not correlate with daily PM10 values. It might be concluded that PM10 monitoring is not appropriate to assess biological effects of PM10 components and their potential health risk. |
